Screening of Media Components to Establish Optimum Conditions for the Differentiation of Hanwoo Adipocytes

Abstract
The current study was conducted to establish optimum conditions for the differentiation of Hanwoo (Korean cattle) adipocytes. Perirenal adipose tissue of Hanwoo was digested with collagenase and obtained stromal-vascular (SV) cells were seeded by 2¡¿10©ùcells/§¢. The cells were maintained with growth media (DMEM/F-12 medium containing 5% FBS, penicillin, streptomycin, and fungizone) at 37¡É under a humidified atmosphere of 95% air : 5% CO©ü. After confluent, media was changed to serum free differentiation media with various growth factors in every 48 hours. Hanwoo adipocytes showed the most differentiated state in morphology and in glycerol-3-phosphate dehydrogenase (GPDH) activity by treatment with media containing 10§¶/§¢ insulin (bovine pancreas), 10§¶/§¢ holo-transferrin (bovine), 0.5mM 1-metyl-3-isobutyl xanthine (MIX), 1ng/§¢ triiodothyronine, 25mM 2,4-thiazolidinedione, 1¥ìM dexamethasone, 25ng/§¢ hydrocortisone, and 1§¡/§¢ lipid mixture (cholesterol rich). Among these growth factors, it seemed that MIX and lipid mixture were essential media components to induce differentiation of Hanwoo adipocytes. Further studies, however. to understand cellular mechanisms underlying lipogenesis in Hanwoo adipocytes are necessary.
